INTENDED USE

Pre-titrated Reagent Kit for human PD-L1 contains calibrated PD-L1 standards and pre-titrated capture and detection antibodies for quantitative measurement of human Pd-L1 in biological samples by enzyme-linked immunoassay (ELISA). An assay protocol is provided with the kit to use the reagents in 96-well ELISA format. It is recommended to dilute all reagents according to this manual. The reagents can also be used with 384-well ELISA plate and optimized for use in other type of immunoassays.

Background

Programmed Death Ligand 1 (PD-L1) is a 40KD transmembrane protein that can link with immune checkpoint protein Programmed Death-1 (PD-1) when T-cell receptors engage with its specific antigen. During T cell receptor signaling, the binding of PD-L1 with PD-1 transmits an inhibitory signals to IL-2 production and T cell proliferation. This inhibitory co-stimulation plays an important role in body’s immune suppressive functions, such as self-tolerance, suppression of immune-rejection during pregnancy and allograft etc. Upregulation of PD-L1 was found to be associated with many cancer types and is believed to contribute to the immune evasion by cancer.
